Cytokines, adhesion molecules, growth factors, antibodies, and other molecules induce and regulate critical cell functions that perpetuate inflammation, leading to tissue injury and clinical phenotype. The nature and intensity of this response as well as the physiological ability to restore homeostasis are to a large extent conditioned by the unique amino acid sequences that define allelic variants on each of the numerous participating mol- ecules. Therefore, the coding genes in their germline configuration play a primary role in determining who is at risk for developing such disorders, how the disease progresses, and how someone responds to therapy. Although genetic components in these diseases are clearly present, the lack of obvious and homogeneous modes of transmission has slowed progress by prevent- ing the full exploitation of classical genetic epidemiologic techniques. Furthermore, autoimmune diseases are characterized by modest disease risk heritability and m- tifaceted interactions with environmental influences. Yet, several recent discoveries have dramatically changed our ability to examine genetic variation as it relates to human disease. In addition to the development of large-scale laboratory methods and tools to efficiently recognize and catalog DNA diversity, over the past few years there has been real progress in the application of new analytical and data-manage- ment approaches.","brand":"WoB","offers":[{"title":"- \/ - \/ INTERNAL","offer_id":52354263810321,"sku":null,"price":0.0,"currency_code":"GBP","in_stock":true},{"title":"GB \/ NEW \/ INGRAM","offer_id":52354267545873,"sku":"NLS9781489997487","price":0.0,"currency_code":"GBP","in_stock":true}],"thumbnail_url":"\/\/cdn.shopify.com\/s\/files\/1\/0784\/4072\/6801\/files\/9781489997487.jpg?v=1758183717"},{"product_id":"oxidative-phosphorylation-in-health-and-disease-book-jan-am-smeitink-9780306482328","title":"Oxidative Phosphorylation in Health and Disease","description":"Mitochondrial diseases are often hard to diagnose. Basic and preclinical studies have revealed the dramatic therapeutic potential of some of these compounds. Thus, preactivation can be used to convert thousands of existing photoactive compounds into new poten- tial anticancer and antiviral agents. Although the process of preactivation uses a principle of photodynamic therapy, the resulting products are strictly chemotherapeutic in nature, in that they function independent of light. Once identified, the photoproducts gener- ated via the process of preactivation can be chemically synthesized. Therapeutic use of light energy by itself or as an activating agent for light- reactive chemicals dates back to ancient Egypt. Since those times, one funda- mental principle of photodynamic therapy has remained constant: light energy is always applied after the light-reactive chemical or photoactive agent has arrived at the target.","brand":"WoB","offers":[{"title":"- \/ - \/ INTERNAL","offer_id":52410328809745,"sku":null,"price":0.0,"currency_code":"GBP","in_stock":true},{"title":"GB \/ NEW \/ INGRAM","offer_id":52410329268497,"sku":"NLS9783662222430","price":0.0,"currency_code":"GBP","in_stock":true}],"thumbnail_url":"\/\/cdn.shopify.com\/s\/files\/1\/0784\/4072\/6801\/files\/9783662222430.jpg?v=1758777542"},{"product_id":"vegf-and-cancer-book-judith-h-harmey-9780306479885","title":"VEGF and Cancer","description":"A comprehensive review of knowledge on the role of vascular endothelial growth factor (VEGF) in cancer. In the following chapters we will review and provide direct evidence for the molecular basis of this group of heterogeneous disorders, which include Rieger syndrome and Rieger anomaly. While ARS patients were initially diagnosed in the early 1930s the genetic basis for ARS was unknown until the recent identification of chromo- somal loci associated with this genetic disorder. In the mid-1990s Drs. Jeffrey C. Murray and Elena V. Semina identified PITX2 through positional cloning tech- niques as a gene associated with ARS. These researchers were able to iden- tify point mutations in PITX2 that were linked with ARS patients. ARS patients presented clinically with several developmental anomalies that fur- ther provided clues about the function of the PITX2 homeobox tran- scription factor. The phenotypic variability of ARS patients indicates that PITX2 can participate with many other faaors to control normal development processes. In the following chapters we will review and provide direct evidence for the molecular basis of this group of heterogeneous disorders, which include Rieger syndrome and Rieger anomaly. While ARS patients were initially diagnosed in the early 1930s the genetic basis for ARS was unknown until the recent identification of chromo- somal loci associated with this genetic disorder. In the mid-1990s Drs. Jeffrey C. Murray and Elena V. Semina identified PITX2 through positional cloning tech- niques as a gene associated with ARS. These researchers were able to iden- tify point mutations in PITX2 that were linked with ARS patients. ARS patients presented clinically with several developmental anomalies that fur- ther provided clues about the function of the PITX2 homeobox tran- scription factor. The phenotypic variability of ARS patients indicates that PITX2 can participate with many other faaors to control normal development processes. The hallmarks of ARS developmental anomalies are eye, tooth and umbilical defects. However, abnormal pituitary, heart, and craniofacial development are also detected. Thus, ARS patients provided the first link of PITX2 involvement in the development of these organs and structures. Biol Cem 1948; 175:315-332). The fact that the chemical structure of our D genome consists of two components that are covalently bound, the genetic information that is replicated by the DNA replication machinery ana DNA methylation that is maintainea by independent enzymatic machinery, has redictably stimulated the imagination and curiosity of generations of mo- Edular biologists. An obvious question was whether DNA methylation was a bearer of additional information to the genetic information and what was the nature of this information? It was tempting to speculate that DNA me- thylation applied some form of control over programming of the genome s expression profile. Once techniques to probe the methylation profile of whole genomes as well as specific genes became available, it became clear that DNA methylation patterns are gene and tissue specific and that patterns of gene expression correlate with patterns of methylation. DNA methylation pat- terns emerged as the only component of the chemical structure of DNA that exhibited tissue and cell specificity. This data seemingly provided an attrac- tively simple explanation for the longstanding dilemma of how could one identical genome manifest itself in so many different forms in multicellular organisms? The DNA methylation pattern has thus become the only known factor to confer upon DNA a unique cellular identity.","brand":"WoB","offers":[{"title":"GB \/ NEW \/ INGRAM","offer_id":52453678842129,"sku":"NLS9781441934161","price":0.0,"currency_code":"GBP","in_stock":true},{"title":"US \/ NEW \/ INGRAM","offer_id":52749501956369,"sku":"NIN9781441934161","price":0.0,"currency_code":"GBP","in_stock":false}],"thumbnail_url":"\/\/cdn.shopify.com\/s\/files\/1\/0784\/4072\/6801\/files\/9781441934161.jpg?v=1759368771"},{"product_id":"hepatitis-delta-virus-book-hiroshi-handa-9780387322308","title":"Hepatitis Delta Virus","description":"Hepatitis Delta Virus is an up-to-date guide to hepatitis D virus (HDV), a human virus with a number of distinctive features. It plays a critical role in immune defence by preventing inflammatory and allergic responses to dietary and non pathogenic microbial antigens. The interest in oral tolerance has been renewed in the recent years, due to novel insights on its cellular mechanisms and potential clinical applications in the treatment of autoimmune diseases.  Oral Tolerance: Cellular and Molecular Basis, Clinical Aspects, and Therapeutic Potential, has been designed as a concise yet comprehensive overview of the newest fundamental and clinical advances in the field. Based on the outstanding contribution of world experts, this book will be helpful to students, clinicians, and researchers working in mucosal immunology and gastroenterology.  The first part of this volume describes the structure and functions of the gastrointestinal mucosa and the fundamental features and mechanisms of oral tolerance, including the role of T cells, cytokines, IgA antibodies, and bacterial antigens. The second part explores the clinical implications of the disruption of oral tolerance in Inflammatory Bowel Diseases, food and milk allergies, and coeliac disease in particular. The final chapter focuses on the clinical potential of oral tolerance as a promising therapeutic tool.","brand":"WoB","offers":[{"title":"- \/ - \/ INTERNAL","offer_id":52480876937489,"sku":null,"price":0.0,"currency_code":"GBP","in_stock":true},{"title":"GB \/ NEW \/ INGRAM","offer_id":52480878346513,"sku":"NLS9781441934253","price":0.0,"currency_code":"GBP","in_stock":true}],"thumbnail_url":"\/\/cdn.shopify.com\/s\/files\/1\/0784\/4072\/6801\/files\/9781441934253.jpg?v=1759850887"},{"product_id":"molecular-pathogenesis-of-cholestasis-book-michael-trauner-9781461347675","title":"Molecular Pathogenesis of Cholestasis","description":"membrane contamination and thus could then be used to study transport function when pre- pared as membrane vesicles. These approaches defined a number of transport functions in the plasma membranes of hepatocyres and ultimately led to the recognition that bile salts and 3 25 other solutes were transported into bile largely by ATP dependent transport mechanisms.2 - The advent ofmolecular cloning techniques and cellular expression systems rapidly acceler- ated progress in this field and led to the molecular characterization ofmost ofthe major mem- brane transport proteins that determine both the hepatic uptake of organic solutes as well as bile salt dependent and bile salt independent canalicular excretion. Overview of the Molecular Mechanisms of Bile Formation The major transporters that determine bile formation are illustrated in Figure 1 and are discussed in greater detail in the subsequent chapters. The enrerohepatic circulation ofbile salts maintains bile salt dependent bile flow. Bile salts are excreted into bile and are largely absorbed in the terminal ileum and then efficiently removed from the portal circulation at the basolateral plasma membrane of the heparocyte. The nature and intensity of this response as well as the physiological ability to restore homeostasis are to a large extent conditioned by the unique amino acid sequences that define allelic variants on each of the numerous participating mol- ecules. Therefore, the coding genes in their germline configuration play a primary role in determining who is at risk for developing such disorders, how the disease progresses, and how someone responds to therapy. Although genetic components in these diseases are clearly present, the lack of obvious and homogeneous modes of transmission has slowed progress by prevent- ing the full exploitation of classical genetic epidemiologic techniques. Furthermore, autoimmune diseases are characterized by modest disease risk heritability and m- tifaceted interactions with environmental influences. Yet, several recent discoveries have dramatically changed our ability to examine genetic variation as it relates to human disease. In addition to the development of large-scale laboratory methods and tools to efficiently recognize and catalog DNA diversity, over the past few years there has been real progress in the application of new analytical and data-manage- ment approaches.","brand":"WoB","offers":[{"title":"GB \/ NEW \/ INGRAM","offer_id":52662995910929,"sku":"NLS9780387360041","price":0.0,"currency_code":"GBP","in_stock":true},{"title":"US \/ NEW \/ INGRAM","offer_id":53397493874961,"sku":"NIN9780387360041","price":0.0,"currency_code":"GBP","in_stock":false}],"thumbnail_url":"\/\/cdn.shopify.com\/s\/files\/1\/0784\/4072\/6801\/files\/9780387360041.jpg?v=1762270870"},{"product_id":"rb-and-tumorigenesis-book-maurizio-fanciulli-9780387321738","title":"Rb and Tumorigenesis","description":"Rb and Tumorigenesis examines how recent advances have demonstrated the interaction of Rb with chromatin remodeling enzymes. Much of this progress can be attributed to our abil- ity to dissect many biological processes at the molecular level. Most spectacular is the technology of molecular biology that allows identi- fication and characterization of genes that participate in the genesis of human cancers. Three major groups of genes appear to play out the drama of cancer development: tumor suppressor genes, mis- match repair genes, and oncogenes. The tumor suppressor genes 1 encode products that are inhibitory to cell proliferation. The loss of these inhibitors, by mutation or deletion, can unleash cells from their restraints to proliferate. Mutations in the mismatch repair 2 10 genes also have been implicated in tumorigenesis. - The inability of cells to repair spontaneously occurring mutations leads to genom- ic instability and could potentially result in the accumulation of car- cinogenic DNA lesions. Finally, activation of proto-oncogenes, which are normal cellular genes, into oncogenes could accelerate the 11 processes of cell proliferation. c-myc was one of the very first proto- oncogenes to be identified and because it normally plays pivotal roles in both cell proliferation and cell death has enticed many to investigate the molecular mechanisms by which it transforms cells.","brand":"WoB","offers":[{"title":"GB \/ NEW \/ INGRAM","offer_id":52665750683921,"sku":"NLS9783662226834","price":0.0,"currency_code":"GBP","in_stock":true}],"thumbnail_url":"\/\/cdn.shopify.com\/s\/files\/1\/0784\/4072\/6801\/files\/9783662226834.jpg?v=1762278135"},{"product_id":"molecular-biology-of-pagets-disease-book-paul-t-sharpe-9783662225073","title":"The Molecular Biology of Paget’s Disease","description":"The osteoclasts in Paget's disease are abnormal both in num- ber and size and show nuclear inclusions as well as cytoplasmic inclusions that suggest a viral etiology for Paget's disease. Use of in vitro model systems for examining osteoclast physiology in Paget's disease has shown that these cells have an increased rate of formation, express high levels of IL-6, IL-6 receptor, and NF-IL-6, as well as most recently, the c-FOS protooncogene. 31 The precur- sors for these pagetic osteoclasts have several abnormalities includ- ing hyperresponsivity to 1,25-dihydroxyvitamin D, presence of 3 measles virus nucleocapsid transcripts, and hyperresponsivity to the marrow microenvironment. In addition, the marrow microenviron- ment is also abnormal in Paget's disease. These data suggest that the initial pathologic event that occurs in Paget's disease affects the osteoclast precursor, as well as the mature osteoclast. The per- sistence of Paget's disease as a highly localized lesion(s) in patients following diagnosis may be due to the abnormalities in the mar- row microenvironment that induce abnormal circulating osteoclast precursors to home to the sites of previous disease and induce increased osteoclast formation in these areas. In areas of normal bone, the normal marrow microenvironment does not induce en- hanced osteoclast formation, but allows these cells to differentiate toward the monocyte macrophage lineage in the form of mature monocytes.","brand":"WoB","offers":[{"title":"- \/ - \/ INTERNAL","offer_id":52671131255057,"sku":null,"price":0.0,"currency_code":"GBP","in_stock":true},{"title":"GB \/ NEW \/ INGRAM","offer_id":52671132107025,"sku":"NLS9783662225073","price":0.0,"currency_code":"GBP","in_stock":true}],"thumbnail_url":"\/\/cdn.shopify.com\/s\/files\/1\/0784\/4072\/6801\/files\/9783662225073.jpg?v=1762291526"}],"url":"https:\/\/www.worldofbooks.com\/en-gb\/collections\/medical-intelligence-unit-book-series.oembed?page=2","provider":"World of Books ","version":"1.0","type":"link"}
