He was awarded a Knox Fellowship to study tumor immunology at Oxford University in the United Kingdom after completing his degree and residency. He returned to the United States to work at the Peter Bent Brigham Hospital, the Sidney Farber Cancer Institute, and the Harvard School of Dental Medicine. Dr. Sonis' research centered on investigations to identify the biology and clinical implications of mucosal toxicities associated with cancer treatments. His research on the molecular and cellular pathogenesis of mucositis has laid the groundwork for a mechanistic model of epithelial injury, as well as therapy options for biological and pharmacological development.
Understanding that genetics has a role in patient risk for mucositis and other toxicities, Dr. Sonis and his colleagues discovered specific canonical pathways that are important in toxicity development and used these as the foundation for gene-based risk prediction models. Dr. Sonis and his colleagues used network theory to describe unique toxicity constellations in chemotherapy patients, building on previous work. Dr. Sonis has a wide range of experience in clinical trial design, implementation, and quality assurance. Many of his past students and residents have gone on to become academic and clinical leaders.
Dr. Sonis has authored numerous papers on the clinical, biological, and health-economic aspects of cancer and its treatment consequences. He has written approximately 200 original articles, reviews, and chapters, as well as seven books and five patents. He has given numerous talks on the clinical and biological aspects of cancer drug toxicity and cancer diagnostics. Dr. Sonis is a founder member of the International Society of Oral Oncology and the International Academy of Oral Oncology, as well as a member of several editorial boards.
